A fully-funded Marie Curie position for PhD student is open at Thermo Fisher Scientific Baltics UAB in Vilnius. Directed evolution-focused discovery research will be carried out in R&D center of the company, while PhD studies will be executed in collaboration with Vilnius University.

PhD topic:

The successful candidate will focus towards design, in vitro evolution and practical applications of artificial DNA clamps. Depending on the polymerase type and the approach identified as a method of choice for enrichment of useful mutants featuring designed artificial DNA clamps, the PhD student will use either compartmentalized self-replication or compartmentalized ribosome display. In the first strategy individual cells will be encapsulated into microfluidics droplets and after release of the cellular content into the compartment those polymerase variants, which are able to amplify their genes under predefined selective pressure, will be accumulated/enriched. In the second scenario in vitro synthesis approach will be explored to make mRNA-ribosome-mutant polymerase complexes for encapsulation. Polymerase mutants will be released from the complex after compartmentalization, and those variants which are able to copy the mRNA molecule within the droplet under conditions of selective pressure will be accumulated/enriched. In each screening stage libraries of selected mutants will be analyzed looking for dynamics of mutation enrichment. The most promising individual mutants will be isolated and their properties characterized. Finally, selected mutants will be tested in field applications where improved properties may be beneficial. At the end of PhD studies we are seeking to have an answer if artificial DNA clamps may be used to improve properties of polymerases.

PhD student will be encouraged to visit our partner laboratories for 3-6 months internship (secondments) at Germany, France, Switzerland or U.K. to gain additional experience. Network-wide training events will include courses and workshops in soft lithography, fabrication and academic writing.

Successful applicants will be funded by the Marie Sklodowska-Curie project “EVOdrops” within the Horizon 2020 program of the European Union, under grant agreement No. 813786. The appointment will be on a temporary basis for a period of 3 years, starting in September 2019.

Eligibility criteria

  1. You must not have resided or carried out your main activity (work, studies, etc.) in Lithuania for more than 12 months in the last 3 years.
  2. You must have a Master or relevant higher education degree in Molecular Biology, Biochemistry or other closely related Life Sciences field.
  3. Your proficiency in the English language should correspond to an ‘English B level’ or higher.
